Ionized boronic acids have a unique affinity to form reversible complexes with 1,2-diols, 1,3-diols, 1,2-hydroxy acids and 1,2­- hydroxylamines. These functionalities are often found in biomolecules and therefore, boronate affinity chromatography is a powerful step in the purification of diverse molecules including carbohydrates, nucleic acids (nucleosides, nucleotides and RNA's), polyphenols (catechols, flavonoids), certain enzymes (serine proteinases) and other proteins. The affinity ligand is bonded onto 6% cross-linked agarose by a highly stable bonding method to provide a robust adsorbent capable of withstanding NaOH cleaning procedures.

Aminobenzamidine is a potent synthetic inhibitor of trypsin-like serine proteases. Immobilized aminobenzamidine has been used in the purification of a large number of serine proteases and esterases by affinity chromatography including trypsin, thrombin, kallikrein, urokinase, acetyl cholinesterase and alkaline phosphatase. Aminobenzamidine Agarose 6XL may also be used in the separation of proteases (for example, trypsin and chymotrypsin) and in the removal of trace protease contamination from your valuable final product. This agarose-based affinity adsorbent offers the superior performance and stability characteristic of all ProMetic's adsorbents.

Properties

Stability

Aminobenzamidine is immobilized to 6% cross-linked agarose via a spacer epoxy linkage that is highly stable over a wide pH range. The aminobenzamidine group degrades at pH's higher than 12, a process accelerated by light. Therefore avoid prolonged exposure to strong alkali solutions and ideally store the adsorbent in the dark below pH 9.

Ligand Content

7.5 +/- 1.0µMol aminobenzamidine per 1ml moist gel.

Capacity

Aminobenzamidine Agarose 6XL binds 25-40mg of trypsin per ml gel depending on the sample condition and pH.

Regeneration

Aminobenzamidine Agarose 6XL can be sanitized before column packing with 70% (v/v) ethanol and can be fully regenerated with 8M urea.

Storage

Aminobenzamidine Agarose 6XL is supplied in 0.1M sodium chloride/ethanol [75/25 (v/v)] and should be stored at 4°C until required. After use, re-equilibrate with preservative and store in the dark at 4°C.

AVIDIN AGAROSE 4XL

Avidin is a basic (pI=10) glycoprotein comprised of four identical subunits having a combined molecular weight of about 67,000 (sub unit molecular weight 15,600). Its interaction with biotin is the strongest known biological interaction between protein and ligand (Ka=1015M-1). The formation of this interaction is rapid, and once formed, it is highly stable and cannot be disrupted by extremes of temperature, pH, organic solvents or denaturing agents. This unique property has provided a valuable tool for development of immunoassays, receptor studies, nucleic acid immobilization, affinity cytochemistry and protein isolation.

For chromatographic applications, the strong interaction of biotin-avidin is a major drawback. Biotinylated proteins are recovered from avidin columns only under denaturing conditions. A number of strategies to overcome this limitation exist. ProMetic has built up a strong expertise in development and manufacture (to GMP/ISO 9001:2000 Quality Standards) of stable agarose adsorbents, which are used in affinity chromatography and protein immobilizations.
