The broadly specific Mimetic™ affinity adsorbents show different binding properties toward any given target protein molecule. These specificity differences are exploited in the PIKSI™ (Protein Isolation Kit for Sorbent Identification) Kit, which has been developed to circumvent the most arduous task of a research project, namely the development of an optimized chromatographic procedure for protein purification, without any prior knowledge of the molecules physio-chemical properties (e.g. iso-electric point, molecular weight, hydrophobicity, ligand specificity).
Designed MIMETIC™ Affinity Ligands - Good Selectivity, Wide Application Range
Affinity chromatography provides a high degree of purification in one step. Although widely used for a number of separations, textile-dye based ligands have not been suitable for large scale purifications. This new generation of computer-modeled synthetic MIMETIC™ ligands offers a range of rationally designed biomimetic structures that interact more specifically with proteins and enzymes. Mimetic™ media easily replace dye-based gels for purification of enzymes and blood proteins, especially those that interact strongly with nucleotides and co-factors.
Improved Stability, Flow Rates and Economy
Mimetic™ media can be reused for many cycles because they can tolerate routine sterilization and regeneration with 1M NaOH. An extremely stable, proprietary bonding chemistry virtually eliminates the ligand leakage problems found with dye-based sorbents. You can also use MIMETIC™ adsorbents at flow rates up to 3 times that of conventional 6% cross-linked agaroses, resulting in increased throughput. Synthetic MIMETIC™ ligands are highly defined and consistent, so you get stability and reproducibility at an economic price. Now you can reduce the cost of your purification with an effective affinity step.
High Capacity and Better Recovery of Activity
The design of the Mimetic™ ligands means higher capacity (up to 50mg protein/ml gel). Mimetic™ ligands minimize loss of activity due to conformational changes, and adsorption and desorption can be carried out under physiological conditions.
Simplify Method Development for Easy Scale-Up
The ultimate goal when purifying proteins is either to provide answers to fundamental questions pertaining to molecular structure and function or to provide bulk commodity products for use in other application (e.g. therapeutics, biotransformations). The development of an optimized chromatographic separation protocol is often the most challenging aspect of such a project due to the molecular diversity of proteins and complexity of biological samples containing the target molecule.
The PIKSI™ M Kit makes fast work out of selecting the optimal MIMETIC™ ligand and conditions. The kit contains ten columns that can be operated simultaneously under gravity flow or individually with a peristaltic pump. Use the kit for method scouting, method development, and then simply scale-up. The adsorbents are manufactured under full GMP and ISO9001:2000 quality systems, and Drug Master Files (DMF) have been filed to make traceability and compliance easier for you. The MIMETIC™ media are available in pack sizes from 25ml to 500L single batches to ensure reliability in any application, from lab scale to large bioprocess.
The PIKSI™ M Kit is supplied with ten packed MIMETIC™ adsorbents and offers the following benefits:
- Rapid screening to identify adsorbents suitable for affinity purification of the target molecule either in the positive mode (binds the target molecule) or negative mode (binds the contaminants).
- The broad specificity of Mimetic™ adsorbents with diverse protein binding properties offers a high level of certainty of identifying a suitable adsorbent in a single experiment.
- Following adsorbent identification binding elution and conditions (pH, ionic strength) can be optimized using the PIKSI™ Kit.
Further method development and small-scale purification may be carried out on our pre-packed 10ml columns or with Mimetic™ adsorbent slurry packed into any conventional chromatography columns.

ProMetics Plasmid DNA Endotoxin Removal Kit is based on Perfluorosorb® S purification media, already proven in large-scale plasmid purification. Perfluorosorb is a highly engineered, polymeric adsorbent that purifies DNA through a reverse phase chromatographic mechanism, and can be applied to primary capture, cccDNA enrichment and endotoxin removal.
Endotoxin levels of < 3EU/mg can be achieved with a single pass rapid procedure of only 7 minutes per prep, from starting levels of > 3000 EU/mg. High DNA capacity of > 3.75 mg and absence of surfactants results in the highest purity preps that will drive your research further.
